site-specific incorporation of 5-methylaminomethyl-2-thiouridine (mnm5s2U) into a model RNA fragment and, together with 2-methyladenosine (m2A), into the native sequence of the Escherichia coli tRNAGlu2 anticodon arm (E. coli ASLGlu2). This approach is also utilized for the synthesis of oligomers modified with multiple 2-thiouridines.
