菊粉简介

菊粉,又名菊糖,是一种由D-果糖经β-(2,1)糖苷键连接而成的链状多糖。其末端常含有一个葡萄糖基。菊粉对人体健康大有裨益:有助于控制血糖水平,适宜于糖尿病患者食用,不引起血糖波动;降低胆固醇量,控制血脂,减小心血管疾病的危害;增殖肠道内的双歧杆菌,预防肠道感染,促进矿物质吸收,防治便秘;在食品工业中,菊粉成功应用于乳制品、面包、糖果、饮料和调味料等领域。例如,其吸湿性强,在食品加工中利用这一特性可延缓水分蒸发,防止产品变味,延长食品的货架期与保质期。

功效与作用

制备

一种耐高温酶法合成菊粉的方法,具体步骤如下:

  1. 培养出芽胞杆菌生产耐高温菊粉合成酶:将发酵原料(蔗糖200g、蛋白胨50g、无机盐2g)经高温灭菌后打入种子罐及发酵罐中。使用出芽胞杆菌TKB28菌株进行摇瓶、种子罐的二级接种,按5%接种量接入发酵罐中,在37℃恒温下发酵16小时左右。随后通过离心将菌体和发酵液分离,并通过分子量20,000的超滤膜浓缩菊粉合成酶溶液,测定所得菊粉合成酶浓缩液的酶活。

  2. 合成菊糖反应:在底物浓度为20%的蔗糖溶液、温度为70℃条件下,按1-1.5U/mL的加入量加入高温菊粉合成酶。初始pH值设置为5.5,在保温8小时后当蔗糖浓度小于5%时,调节pH至7.0,并通过流加蔗糖使蔗糖浓度维持在5-10%,反应进行12小时后停止流加。

  3. 除杂与制粉:反应结束后的溶液通过微滤、离子交换树脂吸附去除杂质。然后,采用干燥法将液体转化为白色粉末状产品,确保其符合质量要求。具体指标包括:
