    Preparation and properties of fluorescent glycosamino-glycuronans labeled with 5-aminofluorescein

    The uronic acid residues of all known glycosaminoglycuronans reacted with 5-aminofluorescein to yield fluorescent glycosaminoglycuronan derivatives, which showed fluorescence characteristics identical to those of fluorescein or 5-acetamidofluorescein. The fluorescent products could be purified by chromatography on Octyl-Sepharose; three preparations of labeled chondroitin 6-sulfate having different degrees of substitution, and a labeled heparin were obtained. Fluorescent hyaluronic acid containing labeled and unlabeled molecules was digested with testicular hyaluronidase to give fluorescent oligosaccharides. Fluorescent chondroitin 6-sulfate was treated with chondroitinase AC to give a nonfluorescent disaccharide and minor proportion of fluorescent octasaccharide. Fluorescent heparin retained its anticoagulant activity, which was similar to that of the starting heparin; its half-life in circulating rabbit blood was 36 min (by fluorometry) and 45 min (by clotting-time assay).

    Hydrogels that degrade under appropriate conditions of pH and temperature by virtue of crosslinking compounds that cleave through an elimination reaction are described. The hydrogels may be used for delivery of various agents, such as pharmaceuticals. This invention provides hydrogels that degrade to smaller, soluble components in a non-enzymatic process upon exposure to physiological conditions and to methods to prepare them. The hydrogels are prepared from crosslinking agents that undergo elimination reactions under physiological conditions, thus cleaving the crosslinking agent from the backbone of the hydrogel. The invention also relates to the crosslinking agents themselves and intermediates in forming the hydro gels of the invention. The biodegradable hydro gels prepared according to the methods of the invention may be of use in diverse fields, including biomedical engineering, absorbent materials, and as carriers for drug delivery.

    A palladium selective chemosensor based on a fluorescein-allyloxy benzene scaffold and a method of detecting palladium ions in a fluid sample with the chemosensor, whereby the fluid sample is contacted with a solution that includes water and the chemosensor to form a mixture. An ultraviolet visible absorption profile and/or a fluorescence emission profile of the mixture is measured to determine a presence or absence of palladium ions in the fluid sample, wherein the chemosensor has an ultraviolet visible absorption peak at 315 to 325 nm and a fluorescence emissions peak at 380 to 400 nm in the solution, and wherein a bathochromic shift in the ultraviolet visible absorption peak to 338 to 342 nm in the mixture and/or a bathochromic shift in the fluorescence emissions peak to 530 to 550 nm in the mixture indicates the presence of palladium ions in the fluid sample.
