A novel linker for the synthesis of C-terminal acetylene-functionalized protected peptides is described. The preparation and application of a new alkyne protecting group is presented. The reactivity and stability of [(3-cyanopropyl)diisopropylsilyl]acetylene is similar to that of the well-known (triisopropylsilyl)acetylene. Its high polarity simplifies chromatographic separation after coupling to aromatics with more than one reactive site. Moreover, compounds with more than one [(3-cyanopropyl)diisopropylsilyl]acetylene group are easily purified after partial deprotection.

Described are compounds of the structure R4-a-Si-(Sp-Y)a-Zb, wherein "a" is integer from 1 to 4; "b" is an integer from 0 to (3 x a); "Z," which is absent when "b "R" or formula (II), wherein each "R" is halogen, C1-6 linear or branched alkyl, alkenyl, or alkynyl or C1-6 linear or branched halo-alkyl, halo-alkenyl, or halo-alkynyl; each "Sp" C1-15 linear or branched alkylenyl or CMS linear or branched halo-alkylenyl; and each "Y" an organic polar group. Also described are electrolyte compositions containing one or more of these compounds.

A new Silyl-based Alkyne Modifying (SAM)-linker for the synthesis of C-terminal acetylene-derivatized peptides is reported. The broad scope of this SAM2-linker is illustrated by manual synthesis of peptides that are side-chain protected, fully deprotected, and disulfide-bridged. Synthesis of a 14-meric (KLAKLAK)(2) derivative by microwave-assisted automated SPPS and a one-pot cleavage click procedure yielding protected 1,2,3-triazole peptide conjugates are also described.
