    A 4-amino(alkyl)cyclohexane-1-carboxamide compound represented by general formula (I), an isomer thereof, and a pharmaceutically acceptable acid addition salt thereof, wherein each symbol is as defined in the specification. These substances have a remarkable, highly persistent and lowly toxic activity of increasing blood stream in coronary, cerebral, renal and peripheral arteries, and are useful as a potent and persistent hypotensive and a drug for preventing and treating diseases of a cardiovascular system such as coronary, cerebral, renal and peripheral arteries. Furthermore, they are useful as an antasthmatic, because they have activities of inhibiting experimental guinea pig asthma caused by the inhalation of histamine and inhibiting contraction of a specimen of an extirpated guinea pig trachea caused by acetylcholine.

    The present invention relates to a medium for direct differentiation of embryonic stem cell-derived mesenchymal stem cells, a method of preparing mesenchymal stem cells by using same, mesenchymal stem cells prepared thereby, and a cell therapy product comprising the same mesenchymal stem cells. In a medium composition and a method according to an embodiment, mesenchymal stem cells may be prepared at high yield within a short period of time. In addition, the method is simple in preparation procedure because of the absence of an embryoid body formation step and allows homogeneous cells to be prepared, thus advantageously providing a cell therapy product within a reduce period of time, compared to conventional methods.

    Disclosed are methods that include separately differentiating stem cells to form cortical neural progenitor cells and differentiating stem cells to form dopaminergic neural progenitor cells. The cortical progenitor cells are transferred to a first compartment of a culture dish and the dopaminergic progenitor cells are transferred to a second compartment of the culture dish, separated by a removable barrier. The cortical progenitor cells and dopaminergic progenitor cells are cultured in medium that promotes differentiation of cortical neurons and dopaminergic neurons, respectively. The barrier is then removed, creating a single chamber containing the cortical progenitor cells and the dopaminergic progenitor cells, separated by a gap. The cortical progenitor cells and the dopaminergic progenitor cells are then cultured in a single medium, which promotes further differentiation of the neurons and also supports formation of connections (such as synapses or synapse-like structures) between at least some of the cortical neurons and dopaminergic neurons.

    Cocktails of chemical inducers of neuron-like properties (CINP) is provided, which includes cAMP agonists, neurogenic small molecules, glycogen synthase kinase inhibitors, TGFβ receptor inhibitors, and BET family bromodomain inhibitors and optionally, a selective inhibitor of ROCK or p38 MAPK. These cocktails are used in a method of inducing neuron-like properties in partially or completely differentiated non-neuronal cells. The method includes contacting cells of a first type (non-neuronal) with the CINPs for a sufficient period of time to result in reprogramming the cell into cells of a second type having neuron-like characteristics (CiNs). Isolated chemically induced neurons (CiNs) can be used in a number of applications, including but not limited to cell therapy.
