检测方法

一种β-NADP二钠盐含量及有关物质(如NAD、ATP、ADP)的测定方法包括以下步骤:

  1. 取sigma对照品beta-烟酰胺腺嘌呤二核苷酸磷酸二钠盐用稀释剂溶解成浓度为1mg/ml的对照品溶液;取样品同样溶解成浓度为0.5mg/ml的供试品溶液。

  3. 分别取sigma对照品NAD、ATP、ADP用流动相溶解成浓度为5ug/ml、2.5ug/ml、2.5ug/ml的杂质对照品溶液;同时取样品用流动相溶解成浓度为0.5mg/ml的样品溶液。
  4. 分别取NAD、ATP、ADP的杂质对照品溶液各20ul,注入液相色谱仪中,记录其峰面积,并按照外标法计算β-烟酰胺腺嘌呤二核苷酸磷酸二钠盐样品中的NAD、ADP和ATP含量。其中,NAD不超过1.0%,ADP不超过0.5%,ATP也应不超过0.5%。
生物活性

Triphosphopyridine nucleotide (NADP) 可在NADP+和NADPH之间交替氧化、切换状态,在很多生物反应中作为电子载体。

用途

烟酰胺腺嘌呤二核苷酸磷酸(NADP)与NADPH形成氧化还原对,是许多辅酶经由电子传输支持的氧化还原反应的关键。尤其是在厌氧反应如脂质和核酸合成中。此外,NADP / NADPH还是细胞色素P450系统和其他氧化酶/还原酶反应系统的耦合辅酶,例如硫氧蛋白还原酶/硫氧蛋白系统的辅酶。
