    The present invention provides methods for purifying and using optically active isomers of erythromycin as well as compositions comprising such optically active isomers. Such optically active isomers having desired actions as an antibiotic substantially separable from undesirable effects on GI motility and the cardiac potassium channels such that the cardiac action potential is not prolonged and the QT interval on the surface EKG (electrocardiogram) is not increased, such that the erythromycin can be useful for more effective therapy of systemic infections. Also disclosed are methods for assaying the levels of such isomers present in the biological fluids.

    A method of entrapping a water-soluble substance in vesicles formed from a siloxane surfactant is carried out by dissolving the substance to be entrapped in water, adding a siloxane surfactant, mildly agitating the mixture and removing excess of water and substance. Further, water-insoluble substances can be entrapped in the vesicles by dissolving the substance to be entrapped in the siloxane surfactant and mildly agitating the substance and the siloxane surfactant. Suitable siloxane surfactants are R-[Si(Me)₂O]₁₄-Si(Me)₂-R in which R is -(CH₂)₃(OCH₂CH₂)₇OH, R-[Si(Me)₂O]₁₄-Si(Me)₂-R in which R is -(CH₂)₃(OCH₂CH₂)₁₂OH, Me₃SiO[Si(Me)₂O]₂₂-[SiMeRO]₂-SiMe₃ in which R is -(CH₂)₃(OCH₂CH₂)₁₂OH and Me₃SiO[Si(Me)₂O]₁₀₃-[SiMeRO]₁₀-SiMe₃ in which R is -(CH₂)₃(OCH₂CH₂)₁₂OH.

    A method of entrapping a water-soluble substance in vesicles formed from a surface active siloxane is carried out by dissolving the substance to be entrapped in water, adding the surface active siloxane, mildly agitating the mixture and removing excess water and substance. Water-insoluble substances are entrapped in the vesicles by dissolving the substance to be entrapped in the surface active siloxane and mildly agitating the substance and the siloxane. The surface active siloxanes essentially consist of tetravalent SiO2 units and monovalent R3SiO1/2 and R'R2SiO1/2 units. The ratio of monovalent units to tetravalent units is from 0.4:1 to 2:1 and from 40 to 90% of all monovalent units are R'R2SiO1/2 units. R is a monovalent hydrocarbon group with up to eight carbon atoms and R' is a polyoxyalkylene group.
