氨基苷类主要作用于细菌体内的核糖体,抑制细菌蛋白质的合成,并破坏细菌细胞膜的完整性。在细胞内,氨基苷类通过依赖能量的Ⅱ期转运系统与核糖体30S亚单位结合,导致错误的蛋白质插入细胞膜,从而使细胞膜渗透性发生改变,细胞内钾离子、腺嘌呤、核苷酸等重要物质外漏,最终导致细菌迅速死亡。进入菌体细胞是一个需氧耗能过程,在缺氧环境下这一过程受到抑制。目前临床使用的氨基苷类在达到治疗浓度(≤25 μg/ml)时可抑制原核细胞合成蛋白质。
