Cytochalasins are known to bind to the barbed, fast growing plus ends of microfilaments, which then blocks both the assembly and disassembly of individual actin monomers from the bound end. Once bound, cytochalasin essentially caps the end of the new actin filament. One cytochalasin will bind to one actin filament. By blocking the polymerization and elongation of actin, cytochalasins can change cellular morphology, inhibit cellular processes such as cell division, and cause cells to undergo apoptosis. 制备方法与用途

简介

细胞松弛素B是肌动蛋白聚合的抑制剂,结合在F-肌动蛋白的正(+)端,阻止其功能。研究者们利用细胞松弛素B、D(Cytochalasin B, CB;Cytochalasin D, CD)或二氢细胞松弛素B(Dihydrocytochalasin B, DCB),对肌动蛋白的功能进行了深入研究,发现肌动蛋白在细胞质中构成微丝,与胞质流动、变形运动、细胞形态的维持、细胞膜的动态变化及细胞器的移动等多种生命活动密切相关。

生物活性

二氢细胞松弛素B(Dihydrocytochalasin B, DCB)是一种细胞分裂抑制剂,能够改变细胞形态,并且其作用与细胞松弛素B相似。值得注意的是,DCB并不抑制葡萄糖转运。它破坏肌动蛋白结构并能逆转地阻断生长因子刺激下的DNA合成能力,从而可逆性地阻止DNA合成的起始过程;此外,DCB还能抑制活性钙的转运。
