of coenzyme A (CoA) is achieved under aqueous conditions using various aliphatic and aromatic carboxylic acids activated as their methyl acylphosphate monoesters. Unlike many hydrophobic activating groups, the anionic methyl acylphosphate mixed anhydride is more compatible with aqueous solvents, making it useful for conducting acylation reactions in an aqueous medium.

The process critically depends on formation of a cross-link between lysyl amino groups of the β-subunits while avoiding reactions with amino groups in the α-subunits. Highly selective acetylation of α-subunit amino groups with methyl acetyl phosphate (MAP) effectively directs subsequent cross-linking to the β-subunits. This outcome leads
