    A dry-type analytical element for immunoassay having at least one water-permeable layer for measuring a ligand in a sample according to enzyme immunoassay, which comprises, (A) a water-soluble macromolecular substrate, and (B) an enzyme conjugated with antibody reacting with the ligand in the sample capable to act on the above water-soluble macromolecular substrate. The above analytical element further comprises, (C) a macromolecular antigen which is a conjugate of the ligand or its derivative with a macromolecular compound in the above water-permeable layer of the above dry-type analytical element. The invention can conduct highly sensitive and simple enzyme immunoassay.

    A dry analytical element for use in detecting a specific component contained in a liquid, which element comprises one or more water permeable layers provided on a water impermeable support, at least one layer of the water-permeable layers containing a composition capable of interacting with said specific component contained in said liquid. The composition contains a leuco dye and a dispersion containing said leuco dye is incorporated in the water-permeable layer, which is formed by dissolving the leuco dye in a hydrophobic solvent containing an aliphatic higher alcohol and dispersing the resulting solution in a hydrophilic medium. The leuco dye has the formula: wherein R¹ represents an unsubstituted or substituted aryl group, R² represents an unsubstituted or substituted alkyl group, and R³ represents an unsubstituted or substituted aryl group. A process for producing said dry analytical element is also disclosed.

    A dry-type analytical element for immunoassay having at least one water-permeable layer for measuring a ligand in a sample according to enzyme immunoassay, which comprises, (A) a water-insoluble macromolecular substrate, and (B) an antibody, reacting with the ligand in the sample, conjugated with an enzyme capable of acting on the above water-insoluble macromolecular substrate. The above analytical substance further comprises, (C) a macromolecular substance which is a conjugate of the ligand or its derivative with a macromolecular compound in the above water-permeable layer of the above dry-type analytical element. The invention can conduct a highly sensitive and simple enzyme immunoassay.

    An integral multilayer analytical element which comprises a first fibrous porous layer, a nonfibrous porous layer, and a second fibrous porous layer superposed in this order on a water-impermeable light-transmissive support. The above three porous layers are integrally laminated to each other substantially closely by an adhesive discontinuously disposed so as to form microspaces continuing through from one layer to the next so as not to interfere with the approximately uniform permeation of liquid. A reagent composition which produces an optically detectable change in the presence of an analyte is incorporated in at least one of said three porous layers. On the other hand, when a nonporous reagent layer is incorporated on the support in the above analytical element, the reagent composition is incorporated in the nonporous reagent layer. By using the analytical element of the invention, the analytical values of various analytes of blood samples can be obtained independently of hematocrit values from whole blood samples and blood plasma samples in the range of the hematocrit values of 0 % to 55 %.

    A dry analysis element for the quantitative analysis of an analyte contained in whole blood comprises a layer exhibiting volume filtration function and a detection layer. The layer exhibiting volume filtration function is made of a cloth selected from microfibrous cloths each having an averagte fiber diameter of from 0.1 to 10 µm or a napped cloth having an average of from 1 to 30 µm. In a first preferred embodiment, the layer exhibiting volume filtration function is composed of a non-woven cloth having a density of from 0.01 to 0.2 g/cm³ and made of fibers each having an average fiber diameter of from 0.2 to 10 µm. In a second preferred embodiment, the layer exhibiting volume filtration function is composed of a napped cloth having a fiber density per cm² of from 7.0 x 10³ to 8.2 x 10⁶, each of the napped fibers having an average diameter of from 1 to 30 microns and an average nap length of 0.5 to 5.0 mm. In a third preferred embodiment, the layer exhibiting volume filtration function is composed of a cloth having a porosity of from 45 to 70% and made of micro filaments each having an average filament diameter of from 0.1 to 7 µm.
