制备方法与用途

简介

嘌呤核苷磷酸化酶简称PNP或PNPase,是嘌呤补救合成途径的关键酶之一,广泛存在于原核和真核生物中。

分类与特点

根据嘌呤核苷磷酸化酶的蛋白结构,可分为两类:低分子量的同源三聚体类和高分子量的同源六聚体类。其中,哺乳动物及部分微生物(如鼠伤寒沙门氏菌、嗜热古菌芝田化叶菌等)的嘌呤核苷磷酸化酶属于同源三聚体类,分子量约为80~100 kDa,每个亚基分子量为30~32 kDa。此类酶通常只能以鸟嘌呤核苷和肌苷作为底物。而同源六聚体类的嘌呤核苷磷酸化酶底物专一性不强,既可接受鸟嘌呤核苷、肌苷,也可腺嘌呤核苷为底物,其分子量约为150 kDa,亚基分子量约为25 kDa。

应用

由于嘌呤核苷磷酸化酶的特殊生物活性,它在医药领域得到了广泛应用:①用于核苷类药物的合成,许多研究表明,核苷类(包括核苷及其类似物)具有广泛的抗肿瘤和抗病毒效果。②用于肿瘤靶向治疗;③用于无机ATP酶活力等的定量测定。
