物化性质

制备方法与用途

概述

磷酸转乙酰基酶(Phosphotransacetylase, PTA)是一种用于测定辅酶A(CoA)效价的酶。辅酶A是由泛酸巯基乙胺腺苷磷酸组成的复合物,通常以氧化型和还原型混合存在,易被氧化。在生物体内,辅酶A是乙酰化酶的辅酶,参与物质代谢过程中的乙酰基传递作用,广泛涉及脂质类、糖类和蛋白质类的代谢,在临床中用于治疗白细胞减少症、原发性血小板减少性紫癜、高脂血症等疾病。目前,辅酶A主要通过微生物发酵法进行酶合成,其效价测定方法主要有磺胺乙酰化酶和磷酸转乙酰基酶(PTA)法两种。

国际上通用的测定方法为PTA法,中国国家标准也采用此法。该方法以还原型CoA单位表示辅酶A的效价。具体原理是,在PTA催化下,乙酰磷酸盐与还原型CoA之间可以可逆地转移乙酰基,形成乙酰辅酶A磷酸
