    The present invention describes compositions and methods for releasing nucleic acids from cells in a form that is suitable for labeling/capture, amplification, or detection in a single reagent addition step. The compositions include a lipid, membrane fluidizing compound, enzyme for degrading cell structure, metal chelators, or one or more nucleic acid probes or primers complementary to the nucleic acid to be detected. The compositions are non-denaturing and non-inhibitory of enzymes or proteins that are used in nucleic acid release, amplification, labeling or detection. The invention also provides kits for performing the above methods.

    The present invention provides a cocktail of reagents for nucleic acid amplification that are stabilized by inclusion of a reversible inhibitor of undesirable reactions. Such cocktail of reagents eliminates the requirement for separate preparation and quality control of each reagent used in a reaction. Methods to prepare stabilized cocktails and to use stabilized cocktails also are included. The stabilized cocktail compositions also can include reagents to release nucleic acid from cells and to label the nucleic acid, allowing detection of nucleic acid in a sample with a single reagent addition step. The invention also provides kits for performing the above methods.

    A dot-blot hybridization method and test kit for detecting normal and mutated genes in unfractionated eucaryotic genomic DNA. Unfractionated single stranded sample DNA is immobilized in a limited zone on a solid support sheet and hybridized with an oligonucleotide probe complementary to the sequence of interest. After stringency washing to remove nonspecifically hybridized probe, the dot-blot zone is examined for the presence of hybridized probe, usually by detection of a label attached to the probe. Hybridization conditions are selected to provide a level of detectable hybridized probe in the dot-blot zone which is readily distinguishable from any background signal associated with the surrounding area on the support sheet. The method eliminates the need for substantial purification and fragmentation of sample DNA required by prior art techniques.

    A method for detecting one or more microorganisms or polynucleotide sequences from eukaroytic sources in a nucleic acid-containing test sample comprising (a) preparing a test sample comprising labeling the nucleic acids in the test sample, (b) preparing one or more process by immobilizing a single-stranded nucleic acid of one or more known microorganisms or sequences from eukaroytic sources, (c) contacting, under hybridization conditions, the labeled single-stranded nucleic acid to form hybridized labeled nucleic acids, and (d) assaying for the hybridized nucleic acids by detecting the label. The method can be used to detect genetic disorders, e.g., sickle-cell anemia.

    A method for detecting (i) one or more microorganisms or (ii) nucleic acid sequences from a prokaryotic source or an eukaryotic source in a nucleic acid-containing test sample comprising (a) labeling the nucleic acids in the test sample, (b) contacting, under hybridization conditions, the labeled hybridizable nucleic acid and one or more immobilized hybridizable nucleic acid probes comprising (i) one or more known microorganisms or (ii) sequences from eukaroytic or prokaryotic sources, to form hybridized labeled nucleic acids, and (d) assaying for the hybridized nucleic acids by detecting the label.
