    苜蓿根瘤咖啡酸O-甲基转移酶的纯化及性质
    摘要:
    摘要 已从苜蓿根瘤中分离并纯化了一种 O-甲基转移酶,该酶以 S-腺苷-1-甲硫氨酸为甲基供体,催化咖啡酸甲基化为阿魏酸。该酶还催化 5-羟基阿魏酸的甲基化。1,6-二氨基己烷琼脂糖 (AH-Sepharose-4B) 与 S-腺苷-1-高半胱氨酸 (SAH) 连接的色谱法得到 35% 的酶活性恢复率。咖啡酸和 S-腺苷-1-甲硫氨酸的 K m 值分别为 58 和 4.1 μM。S -Adenosyl-l -homocysteine 是 S -adenosyl-l -methionine 的有效竞争性抑制剂,K i 为 0.44 μM。通过凝胶过滤色谱法测定的酶的 MW 约为 103 000。
