Thiamet G是一种有效的、选择性O-GlcNAcase抑制剂,Ki值为21 nM,选择性比人溶酶体氨基己糖苷酶高37,000倍。
体外研究在NGF分化的PC-12细胞中,Thiamet G通过抑制O-GlcNAcase而提高O-GlcNAc水平,EC50约为30 nM。100 mM的Thiamet G可以降低tau蛋白在Ser396、Thr231、Ser422 和 Ser262位点的磷酸化程度,分别降低约2.1倍、2.7倍、1.2倍和1.3倍。在肾小球系膜细胞中,Thiamet G(浓度为12.5 nM 和 25 nM)通过增强O-GlcNAc糖基化显著提高p38磷酸化水平。此外,它还作用于功能细胞的O-GlcNAc转移酶或水解酶,修复微管的装配,并部分缓解组蛋白磷酸化。
体内研究以50 mg/kg剂量静脉注射Thiamet G处理大鼠,其能够穿越血脑屏障并增加大脑中的O-GlcNAc水平,这种效应表现出剂量和时间依赖性,同时降低大鼠脑内tau磷酸化。Thiamet G还具有口服生物有效性,并能诱导C57/bl小鼠软骨细胞分化,通过增加分化标记物的基因表达以及MMP-2和-9活性。
