生物碱

生物碱是一类含有氮元素并具有生理活性的天然化合物,大多数具有复杂的氮杂环结构,少数则为非氮杂环有机胺类。它们通常表现出类似碱的行为,在早期曾被译作“膺碱”。尽管某些维生素、氨基酸和肽类等也属于含氮化合物,但习惯上并不归于生物碱范畴。实际上,生物碱至今没有一个严格而确切的定义。

自然界中广泛存在生物碱,尤其在植物界更为常见,动物界也有发现,例如蟾酥中的蟾酥碱(bufotenine)和地龙中的6-氧嘌呤(hypoxanthine)。据统计,在至少140种植物中发现了生物碱,特别是罂粟科、防己科、茄科、豆科、夹竹桃科、毛茛科、小蘖科等。一般来说,双子叶植物含有的生物碱较多,单子叶植物较少,裸子植物和羊齿植物则更少见。含有生物碱的植物通常会包含多种化学结构相似的生物碱。亲缘关系相近的植物所含生物碱的结构往往也较为相似。

生物碱的分类方法多种多样,可按植物来源、生理效应或化学结构进行分类,而后者更为合理。常见的类型包括吡咯类、吡啶类、喹啉类、异喹啉类、吲哚类、咪唑类、嘌呤类、莨菪烷类、萜类、甾体类和有机胺类等。有人将生物碱划分为59种类型,这说明了其结构的复杂性。

除了极少数在室温下为液体的生物碱外,大多数都是结晶型固体,通常无色并具有苦味,具备旋光性和左旋性质。它们呈碱性,这种特性与其分子结构特别是氮原子的结合状态密切相关。季铵类生物碱表现为强碱性,而叔胺和仲胺类生物碱则为中强至弱碱性。某些生物碱分子含有酚基或羧基,因此具有酸碱两性的反应性质。植物中的生物碱通常与有机酸形成盐,或者与糖类缩合生成苷;少数非常弱的生物碱则以游离状态存在。通过碱化处理生物碱盐或水解生物碱苷可获得游离的碱。多数生物碱不溶于水,但易溶于苯、氯仿、乙醚、丙酮和乙醇等有机溶剂中,在稀酸溶液中会形成盐并溶解。

提取生物碱的方法多样,通常先将样品粉碎后用水、稀酸水或乙醇进行提取,然后浓缩提取液并碱化后再用有机溶剂萃取总生物碱。另一种方法是首先碱化样品,再使用有机溶剂提取总生物碱。总碱可采用色谱法或其他方法分离,少数具有挥发性或升华性的生物碱可通过水蒸气蒸馏或升华法制备纯品。

植物体内的生物碱虽然含量较低,但与人类关系密切。人类很早就认识到某些含生物碱的植物或粗提物可用于治病或制毒药。自1806年Serturner从罂粟中分离出吗啡以来,至今已有超过6000种从动植物提取的生物碱被发现,并且其中有近百种已应用于临床。许多生物碱具有治疗价值,如小蘖碱(黄连根茎中的主要成分)具有抗菌消炎作用;利血平来自萝芙木,用于降血压;加兰他敏(石蒜中的一种成分)能缓解小儿麻痹后遗症;吗啡碱(罂粟果皮中含有的著名镇痛剂)、奎宁碱(解热药)、三尖杉碱和长春花碱(抗癌良药)以及秋水仙素(可人工诱导多倍体产生)。某些生物碱还被用作农业杀虫剂。在脊椎动物和无脊椎动物体内也发现了生物碱,其中一些与它们食用的植物有关,例如蟾蜍、蝾螈和某些鱼类中的生物碱被认为是真正的动物代谢产物。
